Conservation and Modification of Epitopes in Polysaccharid Portion of the Lipopolysaccharide of Salmonella typhi


Abstract
Salmonella typhi Ty2 was serially subcultured on the brain heart infusion agar(BHIA) containing 100§¶/ml of acridine orange(AO) to isolate the candidate vaccine strain for the prevention of ty phoid fever. S. typhi AO100, which was subcultured serially on AO-medium 100 times, was exam ined the changes in the antigenic epitopes of lipopolysaccharides(LPSs) using sodium dodecyl sul-fate-polyacrylamide gel electrophoresis and immunoblotting. Although LPSs from S. typhi AO100 had been shown to be composed of higher ratio of long O-side chain LPSs than those of 5. typhi Ty2 in SDS-PAGE, reaction patterns of LPSs from 5. typhi AO100 in immunoblotting were similar to the LPSs of S. typhi Ty2.
These results might indicate the conservation of epitopes in polysaccharide portions of the LPSs from S. typhi AO100 in spite of the serial passages on AO-medium.
